Calkins était surtout connu comme un étudiant de la vie des protozoaires, produisant plusieurs manuels extrêmement influents sur le sujet qui servirent non seulement d'aides à l'enseignement, mais aussi d'importantes déclarations synthétiques dans la science. Ses points de vue ont mûri dans le très cité The Biology of the Protozoa (1926). Son intérêt pour l'ensemble du domaine de la biologie et de la médecine expérimentale a également abouti à un manuel général de biologie en 1914.
